ABSTRACT

Paclitaxel-loaded methoxy poly (ethylene glycol )-b-poly (L-lactic acid) diblock copolymer nanoparticles (PMT) were prepared by a self-emulsification/solvent evaporation method. The PMT morphology, size and its distribution, and drug release in vitro were investigated by DLS, UV, TEM and HPLC. The results indicate that PMT show a spherical morphology with inner core and outer shell. The diameter (nm) of PMT increases with the increase of the drug-loaded amount. The initial burst release is not observed, the drug releasing rate in vitro is lower, and the accumulated release increases with the increase of replacement amout of the pH7. 4 medium. This study develops a new formulation for paclitaxel and provides an experimental basis for the intravenous administration of paclitaxel.

ABSTRACT

Alginate-(Poly-L-Lysine)-Alginate(APA) microcapsules were prepared by Electrostatic Droplet Generator(EDG) technique and the thickness of microcapsule membrane, which was composed by polyelectrolyte complex, were studied in this paper. The theoretical formula was given for the measurement of membrane thickness of APA microcapsules by element analysis of membrane and calculation. The membrane thickness was 7-10 microns by theoretical calculation. On the other hand, the thickness of membrane was measured by SEM and optical microscopy and the results were 7 microns and 12 microns, respectively. The results showed that theoretical calculation is in good accordance with experimental determoination of mermbrane thickness and the membrane thickness of APA microcapsule is about 7-10 microns. The optical microscopy is an easy way to measure membrane thickness.

ABSTRACT

Object To prepare and study a suitable carrier for paclitaxel in water. Methods Cetyl chitosan (CTCS), prepared by reacting chitosan (CS) with chlorocetane under alkaline condition, was soluble and spontaneously formed nanosphere about 100 nm in diameter. And the release in vitro from paclitaxel loaded CTCS nanosphere was measured in phosphate buffer solution (PBS, pH=7 4). Results The balanced release concentration of paclitaxel was deceased and half release time (t 1/2 ) was delayed with the increase of substitution degree of alkyl. Conclusion This kind of nanosphere is an excellent carrier for paclitaxel in water.
